Obec vznikla na mieste pôvodnej osady, ktorú z nej len zvyšky základov stredovekého kostolíka z 12. až 13. storočia. Nová obec pod názvom Rassich sa po prvý raz spomína v roku 1473 už ako mestečko. V roku 1554 dostali Radošovce od uhorského kráľa Ferdinanda I. Trhové a jarmočné právo. V roku 1560 takmer celá obec vyhorela. V priebehu histórie Radošovce patrili k holíčskemu panstvu, od roku 1730 prešli do vlastníctva skalických jezuitov a po zrušení ich rádu sa stali majetkom Habsburgovcov. Obyvateľstvo sa zaoberalo predovšetkým roľníctvom a vinohradníctvom. Doplnkovým zamestnaním boli práce na píle a tkanie konopného plátna. V obci žila početná židovská komunita, ktorá si tu v roku 1830 postavila synagógu.
